In recent years, cancer has become the second cause of death worldwide. Covalent Organic Frameworks (COFs) are new emerging 2 or 3D porous nanomaterials that have attracted tremendous attention in recent years. These materials are composed of light elements including carbon, nitrogen, hydrogen, which are connected with strong covalent bonds. Due to their low density, high surface area and intrinsic porosity, suitable crystallinity and high thermal stability, these materials have become a suitable candidate for many applications such as sensors, drug delivery, separation, catalysts and adsorption. Porosity and high loading capacity have led to highly encapsulation of therapeutic agents into COFs. These materials can modify with different functional groups to achieve targeted and highly biocompatible drug delivery. Also their metal free nature has caused to their proper application in drug delivery and has demonstrated low cytotoxicity in in vitro studies. In this review, firstly different types of COFs and then number of studies conducted with these nanomaterials for drug delivery applications will be reviewed. The last part, the challenges associated with COFs in drug delivery and cancer treatment field will be discussed.
